Genetic engineering is the science of manipulating genetic material of an organism. The concept of genetic engineering was first proposed by Nikolay Timofeev-Ressovsky in 1934. The first artificial genetic modification accomplished using biotechnology was transgenesis, the process of transferring genes from one organism to another, first accomplished by Herbert Boyer and Stanley Cohen in 1973. It was the result of a series of advancements in techniques that allowed the direct modification of the genome. Important advances included the discovery of restriction enzymes and DNA ligases, the ability to design plasmids and technologies like polymerase chain reaction and sequencing. Transformatio…

A genetically modified organism (GMO) is any organism whose genetic material has been altered using genetic engineering techniques. The exact definition of a genetically modified organism and what constitutes genetic engineering varies, with the most common being an organism altered in a way that "does not occur naturally by mating and/or natural recombination". A wide variety of organisms have been genetically modified (GM), including animals, plants, and microorganisms. Genetic modification can include the introduction of new genes or enhancing, altering, or knocking out endogenous genes. In some genetic modifications, genes are transferred within the same species, across species (creatin…

Tinkering with genes and embryos: the multiple invention of transgenic mice c. 1980.. Genetically modified or 'transgenic' mice are a routine experimental tool in biomedical research, commonly produced by injecting DNA into one-cell embryos. These animals were independently invented in 1980 by multiple university groups in the United States and Europe that combined expertise in mouse developmental biology and recombinant DNA techniques, or 'genetic engineering'. In this article, I examine this multiple invention and argue that research strategies, experimental practices, and funding arrangements that led to transgenic mice are best described as tinkering. These creative and speculative ende…

Rudolf Jaenisch (born April 22, 1942) is a professor of biology at the Massachusetts Institute of Technology and a founding member of the Whitehead Institute for Biomedical Research. He is a pioneer of transgenic science, in which an animal's genetic makeup is altered. Jaenisch has focused on creating genetically modified mice to study cancer, epigenetic reprogramming and neurological diseases.

A genetically modified mouse, genetically engineered mouse model (GEMM) or transgenic mouse is a mouse (Mus musculus) that has had its genome altered through the use of genetic engineering techniques. Genetically modified mice are commonly used for research or as animal models of human diseases and are also used for research on genes. Together with patient-derived xenografts (PDXs), GEMMs are the most common in vivo models in cancer research. The two approaches are considered complementary and may be used to recapitulate different aspects of disease. GEMMs are also of great interest for drug development, as they facilitate target validation and the study of response, resistance, toxicity an…

An update on the tools for creating transgenic animal models of human diseases - focus on atherosclerosis.. Animal models of diseases are invaluable tools of modern medicine. More than forty years have passed since the first successful experiments and the spectrum of available models, as well as the list of methods for creating them, have expanded dramatically. The major step forward in creating specific disease models was the development of gene editing techniques, which allowed for targeted modification of the animal's genome. In this review, we discuss the available tools for creating transgenic animal models, such as transgenesis methods, recombinases, and nucleases, including zinc fing…
