However, in terms of nuclei present in skeletal muscle, myocyte nuclei (myonuclei) may be only half of the nuclei present, while nuclei from resident and infiltrating mononuclear cells make up the other half.
Pełniejszy materiał źródłowy / excerpt
Skeletal muscle (commonly referred to as muscle) is one of the three types of vertebrate muscle tissue, the others being cardiac muscle and smooth muscle. They are part of the musculoskeletal system, and typically are attached by tendons to the bones of a skeleton. The skeletal muscle cells (myocytes) are much longer than in the other types of muscle tissue, and are also known as muscle fibers. The tissue of a skeletal muscle is striated – having a striped appearance due to the arrangement of the sarcomeres. A skeletal muscle contains multiple fascicles – bundles of muscle fibers. Each individual fiber and each muscle is surrounded by a type of connective tissue layer of fascia. Muscle fibers are formed from the fusion of developmental myoblasts in a process known as myogenesis resulting in long multinucleated cells. In these cells, the nuclei, termed myonuclei, are located along the inside of the cell membrane. Muscle fibers also have multiple mitochondria to meet energy needs. Muscle fibers are in turn composed of myofibrils. The myofibrils are composed of actin and myosin filaments called myofilaments, repeated in units called sarcomeres, which are the basic functional, contractile units of the muscle fiber necessary for muscle contraction. Muscles are predominantly powered by the oxidation of fats and carbohydrates, but anaerobic chemical reactions are also used, particula…

Spatiotemporal analysis of dystrophin expression during muscle repair.. Background Dystrophin mRNA is produced from a very large genetic locus and transcription of a single mRNA requires approximately 16 h. This prolonged interval between initiation and completion results in unusual transcriptional behaviour: in skeletal muscle, myonuclei express dystrophin continuously and robustly, yet degrade mature transcripts shortly after completion. Consequently, most dystrophin mRNA is nascent, not mature. This implies expression is principally controlled post-transcriptionally, a mechanism that circumvents transcriptional delay, allowing rapid responses to change in demand. Dystrophin protein is ho…

The repair capacity spectrum of human skeletal muscle injury from sports to surgical trauma settings.. Skeletal muscle injury and repair have been a major research focus for more than a century. Muscle injuries are defined by their cause and anatomical location and lie on a spectrum in terms of repair outcomes. From contraction-induced necrosis, which initiates regenerative myogenesis for complete restoration of tissue architecture and function to, at the other end of the spectrum, traumatic volumetric muscle loss (VML), where substantial portions (or the whole) of a muscle are lost, leaving the patient with permanent physical disability. Strain injuries are found between these two extremes…

Aging induces sarcopenia by disrupting the crosstalk between the skeletal muscle microenvironment and myofibers.. Background Sarcopenia is a degenerative muscular disease associated with aging, characterized by a reduction in muscle mass and strength. This disease poses a significant global health challenge, owing to its high prevalence and association with adverse outcomes such as increased frailty, impaired physical function, and elevated mortality risk. A deeper understanding of its underlying mechanisms is urgently warranted for the development of effective therapeutic interventions. Aim of review Skeletal muscle is a heterogeneous tissue composed of various cellular components, includi…

Positioning nuclei at the periphery of skeletal muscle cells.

Endurance training promotes chromatin closure and timely repression of the post-exercise immediate early stress response.. Objectives Endurance training is known to elicit numerous changes in skeletal muscle to enhance performance and function. Many of these adaptations are controlled by the modulation of transcriptional programs in myonuclei. While previous studies have explored alterations in DNA methylation and histone modifications in response to exercise, the specific changes in chromatin restructuring and accessibility, a prerequisite for transcription, are still poorly understood. Methods A multi-omics analysis was performed: ATAC-sequencing was used to map chromatin accessibility in…

Dynamics, specialization and function of muscle nuclei in skeletal muscle.

Cell Death in Skeletal Muscle Diseases: Diverse Roles and Pathological Processes.. Skeletal muscle is vital for movement and metabolism, and its dysfunction underpins disorders like muscular dystrophy and sarcopenia, severely impacting life quality. In these diseases, various cell death pathways are pivotal, driving core pathological features such as fiber loss and chronic inflammation. This study reviews the central role of cell death in skeletal muscle diseases, and analyzes its roles and mechanisms in genetic muscle disorders such as Duchenne muscular dystrophy (DMD), glycogen storage diseases (GSD), mitochondrial myopathies, as well as acquired muscle disorders such as idiopathic inflam…
