Aging in dogs varies from breed to breed, and affects the dog's health and physical ability. As with humans, advanced years often bring changes in a dog's ability to hear, see, and move about easily. Skin condition, appetite, and energy levels often degrade with geriatric age. Medical conditions such as cancer, kidney failure, arthritis, dementia, and joint conditions, and other signs of old age may appear. The aging profile of dogs varies according to their adult size (often determined by their breed): smaller breeds have an average lifespan of 10–15 years, with some even exceeding 18 years in age; medium breeds typically live for 10 to 13 years; and giant dog breeds have the lowest minimu…

Pedigree structure, inbreeding, and survival analysis in an Irish Wolfhound cohort.. Background The Irish Wolfhound (IW) is a giant dog breed characterized by a relatively short lifespan, a high prevalence of heritable disorders, and extensive relatedness among lineages. Despite the recognized importance of inbreeding and population structure in purebred dogs, relatively little is known about how pedigree structure and demographic parameters influence survival patterns in large multinational IW populations. The aim of this study was to investigate pedigree structure, inbreeding, demographic parameters, and survival patterns in a multinational cohort of Irish Wolfhounds. An integrated pedigr…

Computational prescreening of anticancer drugs prior to in vitro validation, primarily in dogs with exploratory cat data, based on cross-species binding conservation.. Advances in veterinary medicine have prolonged the lifespan of companion animals. As a result, the burden of neoplastic diseases has increased. While targeted anticancer drugs are widely used in human oncology, their clinical translation to companion animals remains limited. Therefore, a systematic framework for prioritizing candidate drugs before in vitro or in vivo testing would be a valuable strategy in veterinary medicine. In the present study, we evaluated the cross-species binding potential of FDA-approved targeted anti…

How can we achieve more accurate reporting of average dog lifespan?. Abstract Despite major advances in our understanding of dogs as a biological system (including genetics/epigenetics, physiology, cognition, and behavior), the veterinary field lacks consensus around a critical piece of information: namely, the average lifespan/life expectancy of a domestic dog. This deficiency is due in part to unavailable and/or inconsistent collection of dog mortality data. In an effort to review historical and current reports of dog lifespan to determine whether the domestic dog’s lifespan has changed over time, we found that incongruous data were prohibitive to conducting a formal meta-analysis of dog…

Genotypic and phenotypic consequences of domestication in dogs.. Runs of homozygosity (ROH) are genomic regions that arise when identical haplotypes are inherited from a shared ancestor. In this study, we explored ROH across 556 whole-genome sequences from domesticated and non-domesticated dogs. Then, we leveraged ROH from 466 breed dogs, representing 13 breed groups and 13 phenotypic traits, to investigate associations between genetic diversity and non-disease phenotypes. We identified significant associations between the ROH-based inbreeding coefficient (FROH) and multiple phenotypes. These include three quantitative traits (height, weight, lifespan) and ten morphological and coat-related…

The cross-sectional average inequality in lifespan (CAL†): A lifespan variation measure that reflects the mortality histories of cohorts. Lifespan variation is a key metric of mortality that describes both individual uncertaintyabout the length of life and heterogeneity in population health. We propose a novel andtimely lifespan variation measure, which we call the Cross-sectional Average Inequality in Lifespan. This new index provides an alternative perspective on the analysis of lifespan inequality by combining the mortality histories of all cohorts present in a cross-sectional approach. We demonstrate how differences in the Cross-sectional Average Inequality in Lifespan measure can be de…
